Mechanism that maintains stem cells readiness identified

ScienceDaily (May 31, 2012) ? An immune-system receptor plays an unexpected but crucially important role in keeping stem cells from differentiating and in helping blood cancer cells grow, researchers at UT Southwestern Medical Center report today in the journal Nature.

"Cancer cells grow rapidly in part because they fail to differentiate into mature cells. Drugs that induce differentiation can be used to treat cancers," said Dr. Chengcheng "Alec" Zhang, assistant professor in UT Southwestern's departments of physiology and developmental biology. "Our research identified a protein receptor on cancer cells that inhibits differentiation, and knowing the identity of this protein should facilitate the development of new drugs to treat cancers."

The family of proteins investigated in the study could help open a new field of biology integrating immunology with stem cell and cancer research, he added.

"The receptor we identified turned out to be a protein called a classical immune inhibitory receptor, which is known to maintain stemness of normal adult stem cells and to be important in the development of leukemia," he said.

Stemness refers to the blood stem cells' potential to develop into a range of different kinds of cells as needed, for instance to replenish red blood cells lost to bleeding or to produce more white blood cells to fight off infection. Once stem cells differentiate into adult cells, they cannot go back to being stem cells. Current thinking is that the body has a finite number of stem cells and it is best to avoid depleting them, Dr. Zhang explained.

Prior to this study, no high-affinity receptors had been identified for the family of seven proteins called the human angiopoetic-like proteins. These seven proteins are known to be involved in inflammation, supporting the activity of stem cells, breaking down fats in the blood, and growing new blood vessels to nourish tumors. Because the receptor to which these proteins bind had not been identified, the angiopoetic-like proteins were referred to as "orphans," he said.

The researchers found that the human immune-inhibitory receptor LILRB2 and a corresponding receptor on the surface of mouse cells bind to several of the angiopoetic-like proteins. Further studies, Dr. Zhang said, showed that two of the seven family members bind particularly well to the LILRB2 receptor and that binding exerts an inhibitory effect on the cell, similar to a car's brakes.

In the case of stem cells, inhibition keeps them in their stem state. They retain their potential to mature into all kinds of blood cells as needed but they don't use up their energy differentiating into mature cells. That inhibition helps stem cells maintain their potential to create new stem cells because in addition to differentiation, self-renewal is the cells' other major activity, Dr. Zhang said. He stressed that the inhibition doesn't cause them to create new stem cells but does preserve their potential to do so.

In future research, the scientists hope to find subtle differences between stem cells and leukemia cells that will identify treatments to block the receptors' action only in leukemia.

Other UT Southwestern researchers involved in the study from the departments of physiology and developmental biology include postdoctoral researchers Dr. ChangHao Cui, Dr. Xiaoli Chen, Dr. Chaozheng Zhang, Dr. HoangDinh Huynh, and Dr. Xunlei Kang; senior research associates Robert Silvany and Jiyuan Li; and graduate student Xuan Wan. Researchers from the department of immunology include former technician Alberto Puig Cant? and Dr. E. Sally Ward, professor of immunology.

Needle-free device could deliver powdered drugs

"You'll just feel a sharp sting..." No, don't look away, because these words may soon be a thing of the past. A needleless device that jets medicine through the skin could bring an end to painful injections. It could also provide developing countries with a way to deliver powdered drugs, which do not require refrigeration.

There are several needleless systems already on the market, which use explosive forces to eject a liquid drug through a narrow opening at pressures high enough to penetrate the skin. However, they are designed to eject the drug in an "all or nothing" fashion, releasing the same amount of medicine to the same depth every time.

To counter this limitation, Ian Hunter and Nora Hogan at the Massachusetts Institute of Technology, together with Andrew Taberner at the University of Auckland, New Zealand, developed a device based on a "Lorentz force" mechanism.

This consists of a coiled wire that is wrapped around a powerful magnet at one end and attached to a piston at the other. When a current is applied to the wire it interacts with the magnet, pushing the piston forwards. This in turn pushes a liquid drug through a hole the width of a mosquito's proboscis and into the skin. The force can be controlled by varying the current applied to the coil, which allows precise control over dose and depth.

Targeted drugs

Because existing needleless devices offer less control they generally deliver less of the drug to the target site. In some cases, say Hunter's team, just 10 per cent of the drug reaches the correct tissue. Their tests in animal tissue showed that the new device delivers more than 80 per cent of the drug to the target.

The team is now working on adding a vibratory mechanism to their device to allow it to transform powdered drugs into a form suitable for injection. "If you vibrate a powder the drug crystals jiggle around, become energised and behave in a similar way to a fluid," says Hunter.

If successful, their device could help increase the availability of drugs in the developing world, because powdered drugs are more stable than liquid drugs at room temperature ? liquid drugs must be refrigerated to keep them stable. "It's early days but we're pretty excited," says Hunter.

WASHINGTON (AP) ? Republican presidential candidate Mitt Romney is calling for more "assertive measures" against Syrian President Bashar Assad, including arming the Syrian opposition. The Obama administration so far has declined to help arm the opposition.

Romney said in a written statement that he welcomed Tuesday's mass expulsion of Syrian diplomats by the U.S. and other countries to protest the weekend massacre of dozens of civilians, including children.

But he said the move underscores the need for stronger measures to end Assad's regime. He said President Barack Obama's "lack of leadership" had created a "policy of paralysis" that has led to the killing of an estimated 10,000 Syrians.

Romney called for increasing pressure on Russia to stop selling arms to Syria, and for Russia to end its obstruction at the United Nations.

U.N. Security Council condemns Syria over massacre

AMMAN/UNITED NATIONS (Reuters) - The U.N. Security Council on Sunday unanimously condemned the killing of at least 108 people, including many children, in the Syrian town of Houla, a sign of mounting outrage at the massacre that the government and rebels blamed on each other.

Images of bloodied and lifeless young bodies, laid carefully side by side after the onslaught on Friday, triggered shock around the world and underlined the failure of a six-week-old U.N. ceasefire plan to stop the violence.

Western and Arab states opposed to Syrian President Bashar al-Assad put the blame for the deaths squarely on the government. But Damascus rejected the charge.

"The Security Council condemned in the strongest possible terms the killings, confirmed by United Nations observers, of dozens of men, women and children and the wounding of hundreds more in the village of (Houla), near Homs, in attacks that involved a series of government artillery and tank shellings on a residential neighborhood," the non-binding statement said.

"Such outrageous use of force against civilian population constitutes a violation of applicable international law and of the commitments of the Syrian Government under United Nations Security Council Resolutions," the statement said.

The United Nations believes that at least 108 people were killed in Houla, U.N. peacekeeping chief Herve Ladsous said.

Russia, which along with China has vetoed two Security Council resolutions calling for tougher action against Damascus, said the "tragic" events in Syria deserve condemnation and called for a U.N. assessment of the violence there.

Russian Deputy U.N. Ambassador Alexander Pankin said the circumstances surrounding the massacre were "murky" and rejected the idea that the evidence clearly showed Damascus was guilty.

The head of the U.N. observer force, General Robert Mood, briefed the council by video link. Pankin said Mood "did not link directly the (army's) shelling with numbers of deaths."

U.N. Secretary-General Ban Ki-moon sent the council a letter that appeared to give ammunition to both sides.

He said the observers "viewed the bodies of the dead and confirmed from an examination of ordnance that artillery and tank shells were fired at a residential neighborhood." The rebels do not have artillery and tanks.

But Ban also said U.N. monitors observed shotgun wounds on some of the bodies, which could indicate close-range attacks by rebels, as Pankin suggested, or could be the result of follow-up attacks by the army after it stopped shelling.

"While the detailed circumstances are unknown, we can confirm that there has been artillery and mortar shelling," Ban said. "There have also been other forms of violence, including shootings at close range and severe physical abuse."

International mediator Kofi Annan and Ladsous are expected to brief the council on Syria on Wednesday.

RUSSIA OPPOSES SANCTIONS

British Ambassador Mark Lyall Grant did not have any doubts about who was responsible for the events in Houla.

"It seems quite clear that the massacre in Houla was caused by heavy bombardment, by government artillery and tanks," Lyall Grant said. After the council meeting he said it was time for the council to discuss "next steps" - a code word for sanctions.

"The fact is, it is an atrocity and it was perpetrated by the Syrian government," Lyall Grant said.

Russia, however, rejects the idea of sanctioning its ally and has accused the United States and Europe of pursuing Libya-style regime change in Syria, where Assad has been trying to crush a 14-month-old insurgency that began peacefully but has become increasingly militarized.

Syria is home to the Russian navy's only warm-water port outside of the former Soviet Union and is a major purchaser of Russian weapons.

Syrian Ambassador Bashar Ja'afari reiterated his government's denial, saying the massacre was committed by "armed terrorist groups" - the Syrian government's term for the rebels. He also dismissed the "tsunami of lies" of the British, French and German envoys, who blamed the government for the massacre.

"Women, children and old men were shot dead. This is not the hallmark of the heroic Syrian army," Foreign Ministry spokesman Jihad Makdesi told reporters in Damascus.

The Houla massacre is among the worst carnage in the uprising against Assad, which has cost about 10,000 lives.

In his public comments, Mood has called the killings "a very tragical expression" of the situation in Syria, but refrained from apportioning blame.

"For myself, I have had patrols on the ground all the day yesterday afternoon and today we are gathering facts on the ground and then we will draw our own conclusions," Mood told the BBC in a telephone interview on Sunday.

But Ban and Annan, the U.N. and Arab League envoy for Syria, accused the Syrian government of using artillery in populated areas.

"This appalling and brutal crime involving indiscriminate and disproportionate use of force is a flagrant violation of international law and of the commitments of the Syrian Government to cease the use of heavy weapons in population centers and violence in all its forms," they said in a joint statement on Saturday.

Annan is planning to visit Damascus soon. Ja'afari suggested Annan could arrive as early as Monday.

Russia's Pankin said that whoever caused the massacre wanted to disrupt Annan's visit. "We don't believe the Syrian government would be interested in spoiling the visit of (Annan) ... by doing something like that."

"CORPSES WERE PILED ON TOP OF EACH OTHER"

Opposition activists said Assad's forces shelled Houla after a protest and then clashed with fighters from the Sunni Muslim-led insurgency.

Activists say Assad's "shabbiha" militia, loyal to an establishment dominated by members of the minority Alawite sect, then hacked dozens of the victims to death, or shot them.

Maysara al-Hilawi said he saw the bodies of six children and their parents in a ransacked house in the town.

"The Abdelrazzak family house was the first one I entered. The children's corpses were piled on top of each other, either with their throats cut or shot at close range," Hilawi, an opposition activist, said by telephone from the area.

A video distributed by activists showed an injured woman, who said she had survived the massacre, blaming shabbiha militiamen for the carnage.

"They entered our homes ... men wearing fatigues herding us like sheep in the room and started spraying bullets at us," the woman said. "My father died and my brother, my mother's only son. Seven sisters were killed," the woman said lying next to another injured woman and near a baby with a chest wound.

The White House said it was horrified by credible reports of brutal attacks on women and children in Houla.

"These acts serve as a vile testament to an illegitimate regime that responds to peaceful political protest with unspeakable and inhuman brutality," a White House spokesman said.

The Gulf Cooperation Council of Sunni-led monarchies accused Assad's soldiers of using excessive force and urged the international community to "assume its responsibilities to halt the daily bloodshed in Syria."

Normally a minority shareholder is a shareholder who has less than 50% of the outstanding shares of a company with the majority shareholder(s) having over 50% of the outstanding shares. The size of the company (closely held company v. publically traded company) usually determines the voting power of the shareholders and how the minority interest is actually affected. Publically traded corporations normally have numerous shareholders who may combine their voting interest to control how the company is run, whereas closely held corporations usually have very few shareholders. Often times, closely held corporations have one or two majority shareholders who in turn control how the company is run, thereby suppressing the rights of the minority shareholder(s).

Unless otherwise stated in the articles of incorporation or bylaws of the company, the majority rule is used regarding any decision brought before shareholders, except the appointment of the board of directors. La. R.S. art. 12:75(I). Therefore, unless the articles or bylaws allow for cumulative voting or some other form of minority voting protection, a minority shareholder will be at the mercy of the majority shareholder when it comes to making decisions for the company or electing directors and officers to make decisions for the company. As one may suspect, this control can cause a serious rift between the shareholders which often leads to a falling out.

However, Louisiana law offers some remedy for minority shareholders who feel they are being treated unfairly. One of the more gutsy options for a minority shareholder is to file a petition for involuntary dissolution of the company. La. R.S. art. 12:143. A minority shareholder may have grounds for involuntary dissolution if can prove that the directors or officers of the corporation have been guilty of gross and persistent ultra vires acts and thereby breached their fiduciary duty to the company. However, this type of action carries a very serious burden of proof and courts normally do not find directors guilty of ultra vires actions. One specific case did rule in favor of a minority shareholder and force liquidation of the company whereby the minority shareholder proved that the majority shareholders, who were also serving as the directors of the company, had conducted ultra vires activities. Gooding v. Millet, 430 So.2d 742 (La. App. 5th Cir. 1978). The court in Gooding determined that the corporate officers were guilty of gross and persistent ultra vires actions when the officers failed to issue stock certificates, failed to hold annual meetings, failed to keep adequate financial records and granted themselves salaries without a proper vote at a shareholders meeting.

Another option for a minority shareholder to flex their muscle is through a writ of mandamus. A writ of mandamus directs a corporate officer to perform their required duties. La. C.C.P. art. 3861. These required duties include the recognition of rights of the shareholders or some other obligation required according to the bylaws of the corporation. La. C.C.P. art. 3864. One of the more common mandamus actions is the shareholders' ability to inspect financial records of the corporation, a right that courts often recognize in Louisiana. Feil v. Greater Lakeside Corporation, 31 So.2d 520 (La. App. 5th Cir. 1/26/10). A mandamus action lets the controlling shareholder(s) know that the minority shareholder is aware of their rights and is not afraid of using the legal system to enforce those rights.

The above mentioned remedies are just a couple of examples of the rights provided to minority shareholders under Louisiana law. While being a minority shareholder can be frustrating at times, it is important to remember that you do have options.
